Endocrine Disruption in Practice: From EAS-Modalities to Weight-of-Evidence Strategies

SUMMARY

The upcoming presentation, "Endocrine Disruption in Practice: From EAS-Modalities to Weight-of-Evidence Strategies" delivers a strategic overview of European regulatory frameworks and testing methodologies for Endocrine Disruptor (ED) identification. It outlines the three mandatory pillars required by ECHA and EFSA to formally classify a substance as an ED - establishing Endocrine Activity, an Adverse Effect, and a biologically plausible Mode of Action connecting the two. Focusing specifically on Estrogen, Androgen, and Steroidogenesis (EAS) modalities, the overview charts the OECD testing hierarchy from Level 2 in vitro mechanisms to Level 3 in vivo screens.

Crucially, the session highlights the heavy operational burdens associated with the OECD 443 gold standard and introduces strategic alternatives. By leveraging Weight of Evidence (WoE) rationales, QSAR computational modeling, and New Approach Methodologies (NAMs), toxicologists can build scientifically sound arguments to support testing waivers and successfully navigate complex regulatory outcomes.

Giada MARCHETTI

Toxicologist at Staphyt, Giada is a specialist in regulatory compliance, particularly on EU-REACH and plant protection products (PPP) area. She manages study monitoring, health risk assessments, CLP classification, and expert statements. She brings solid expertise in QSAR and read-across, having even participated, in her previous roles, in the development of new predictive models for the toxicological properties of complex substances. Giada is also a major asset to the team on the topic of Endocrine Disruptors (ED), for which she conducts assessments and drafts expert opinions.

Constança Pais Do Amaral

Constança is a toxicologist at Staphyt, specialist in plant protection products (PPP), with a PhD in Biomedical Sciences. She is involved in study monitoring, human health/non-dietary risk assessments, CLP toxicological classification and literature search, and develops skills in biocide and REACH regulation. She brings a strong scientific foundation and hand-on regulatory experience to toxicological challenges. Constança also plays an active role in Endocrine Disruptors (ED) assessments, supporting clients across industries.
